Aspergillus oryzae and related koji molds

Koji spores, or tane-koji, are the inoculum used to begin a fresh batch of koji. They are sold either as a concentrated powder or on dried grains of rice or barley; both forms can inoculate a different cooked-grain substrate.12

Commercial tane-koji is not just raw mold dust. Traditional producers select strains for the enzyme profile they want, especially the amylases, proteases, and lipases that make koji useful for starch, protein, and fat breakdown. In dry form the spores keep for months, which is one reason they function so well as a pantry ingredient rather than a strictly perishable culture.2

Once those spores land on a suitable cooked grain, they germinate into hyphae, spread into a white mycelial network, and turn the grain into sweet, fragrant koji over roughly two days.3 The mold’s food value is enzymatic: it breaks large starch and protein molecules into smaller sugars, peptides, and amino acids that later fermentations can use. Japanese agricultural standards describe koji mold in those terms and distinguish Aspergillus oryzae from related strains used for soy sauce and other ferments.4

When using the powdered form, it helps to dilute a very small dose with toasted flour or starch first so it can be sifted evenly across the substrate; otherwise it is easy to overconcentrate one patch and miss another.1 The distinction between a selected food strain and an unknown environmental mold matters: commercial koji starters are chosen for stable growth and useful enzyme production, while wild mold on cooked grain is not an equivalent ingredient. That is why spores belong in the pantry as a named culture with a known identity, not as a generic invitation to let any mold colonize food.
